鴿,一種十分常見的鳥,世界各地廣泛飼養,對于鴿子的繁殖,繁殖效率不掌握方法容易低迷,繁殖速度較慢,如何才能提高?其實鴿子繁殖的影響很多,但是主要還是兩個問題,一個是健康的問題,一個是性別的問題;這兩個問題就算是非常注意有時候也是不可避免的。

鴿子是一種單態鳥,并且是典型的一夫一妻制,一夫一妻制的配偶性使得成鴿對配偶是有選擇的,一旦配偶后,公母鴿總是親密地生活在一起,共同承擔筑巢、孵卵、哺育乳鴿、守衛巢窩等職責;這給人工繁殖帶來了很大的困難,對鴿子羽毛進行研磨提取DNA來鑒定性別和基因優良與否,對動物無害,且準確,在管理上有較廣闊前景。

對于鴿子羽毛的研磨,現有技術中羽毛研磨流程工藝有:清洗、烘干、膨化、研磨等加工步驟,而在研磨這一塊基本是直接采用常用粉碎機對膨化后的羽毛進行粉碎,無法對原羽毛進行徹底粉碎且無法達到能夠提取鴿子DNA的研磨成品質量。

對于許多室溫下過于柔軟或敏感的分析樣品, 在常規的粉碎過程中,易裂解成多種形式;所以冷卻技術便成為了保存樣品完整性的關鍵手段,一旦樣品被浸沒在液氮中進行冷卻時,會迅速變脆,從而更容易被粉碎, 粉碎時間也相對較少, 關鍵是在低溫下,一些樣品中的結構和組成信息可以得到有效保留。

這些樣品包括聚合物、橡膠、紡織原料、谷物顆粒、頭發、指甲、皮膚、骨頭、肌肉組織等等; 低溫粉碎已經是化學分析中的重要的預處理手段。
研磨實例:
一: 選擇合適大小的離心管;
二: 加入1顆研磨珠,加入樣品,放入液氮中浸泡一段時間;
三:按照要求設置好相關參數;
四:取出離心管,加入提取液做后續實驗。
